5,7,3',4',5'-五甲氧基黄酮对K562细胞增殖和凋亡的影响Effect of 5,7,3',4',5'-pentamethoxyflavone on the apoptosis of K562 cells

摘要(Abstract):
目的:研究5,7,3',4',5'-五甲氧基黄酮(5,7,3',4',5'-pentamethoxyflavone,PMF)对人慢性粒细胞白血病细胞株K562细胞凋亡的影响。方法:对K562细胞进行常规培养,将其分为正常组、低剂量组、中剂量组、高剂量组,分别采用对应剂量的PMF进行处理。使用四甲基偶氮唑盐(MTT)法检测PMF对K562细胞存活率的影响,采用吖啶橙(Acridine Orange,AO)染色法观察PMF对K562细胞凋亡的影响,采用蛋白免疫印迹法(Western Blot)检测PMF处理后,K562细胞Bcl-2、caspase-3的蛋白表达水平。结果:MTT实验的结果显示,PMF对K562细胞有一定的抑制其增殖的作用,随着PMF剂量的增加,对K562细胞的增殖抑制作用越强(P<0.05)。蛋白免疫印迹法结果显示,随着PMF剂量的增加,与凋亡相关的蛋白Bcl-2的表达下调,caspase-3的表达明显比正常组高并且逐步上调。吖啶橙(AO)染色后可明显见到凋亡的细胞随PMF剂量的增加越来越多。结论:PMF可以抑制K562细胞的增殖,并通过促进caspase-3蛋白的表达和抑制Bcl-2蛋白表达,促进K562细胞凋亡。
